Objective To investigate the effect of photodynamic therapy combined with ciprofloxacin in the treatment of seawater immersion injury. Methods A total of 50 BALB/c mice were randomly divided into five groups, with 10 mice in each group: a clean wound control group(Control group), a seawater immersion injury model group (SII group), a seawater immersion injury with photodynamic therapy group (PDT group), a seawater immersion injury with ciprofloxacin treatment group (CPFX group), and a seawater immersion injury with photodynamic therapy combined with ciprofloxacin treatment group (PDT+CPFX group). The mice in the PDT group underwent a photodynamic therapy of 20 minutes (power density: 15 mW/cm2), the mice in the CPFX group received ciprofloxacin ointment on the wound surface every 24 hours for 7 days, and photodynamic therapy was administered to the PDT+CPFX group for 20 minutes, followed by daily topical application of ciprofloxacin ointment for 7 consecutive days. Wound healing was monitored by photographing the wounds on days 0, 3, 7, 10, and 14 of the experiment. Hematoxylin and Eosin (HE) staining was used to observe the pathological changes of the wounds in each group on days 3, 7, and 10, and to observe whether photodynamic therapy would cause pathological changes in the heart, liver, spleen, lung, kidney and brain tissues of each group of mice. Immunohistochemistry (IHC) was used to detect the levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in in wound of each group of mice on days 3, 7, and 10 of the experiment. Results Compared with the Control group, the SII group showed delayed wound healing, significant inflammatory infiltration, and increased levels of TNF-α and IL-6 (P<0.05). Compared with the SII group, the PDT+CPFX group showed the best wound healing, with significantly better epidermal regeneration and fewer inflammatory cell infiltrates on days 10 and 14 (P<0.05). Photodynamic therapy did not cause significant pathological changes in the heart, liver, spleen, lungs, kidneys or brain of the mice. Conclusions Photodynamic therapy combined with ciprofloxacin synergistically promotes wound healing in seawater immersion injuries.
