Association of IL-6 gene promoter region -643C/G polymorphism with diabetic retinopathy

Abstract

Objective To investigate the association of IL-6 gene promoter region -643C/G polymorphism with diabetic retinopathy. Methods The polymorphism of IL-6 gene promoter region -634 C/G in 245 unrelated Guangdong Han people with type 2 diabetes ,including 153 cases without diabetic retinopathy (DR0 group) ,51 cases without proliferative diabetic retinopathy (NPDR)(DR1 group), 41 cases with proliferative diabetic retinopathy (PDR) (DR2 group), and 101 normal controls(NC group) were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods. The genotype frequency, allele frequency and relative clinical data were compared between groups. Results (1)There were no significant differences in the clinical characteristics except for diabetic duration among DR0, DR1, DR2 groups. (2) The frequencies of IL-6-643 G/G genotype and G allele frequencies among DR0 DR1 DR2 groups were statistically different. There were significantly different in GG genotype between DR0 and DR group,comparison between any two means were not significant differences in G allele frequencies.(3) Logistic regression analysis showed that IL-6 gene promoter region-643G/G genotype,fasting plasma glucose and diabetic duration were risk factors of DR. Conclusions The IL-6 promoter region-643G/G genotype may be a genetic risk factor of DR development.
