Effects of sodium selenite on T lymphocyte proliferation and mitochondrial function in vitro

Effects of sodium selenite on T lymphocyte proliferation and mitochondrial function in vitro

Abstract

Objective To study whether or not there is protective effect of sodium selenite on T lymphocytes of heroin addicts, and if it correlates with mitochondrial uncoupling related indicators. Methods T lymphocytes extracted from peripheral blood of people who abuse heroin in different periods. Cell proliferation was detected by MTT, the levels of ROS, △Ψm were detected by fluorescence spectrophotometer, MPTP was measured by ultraviolet spectrophotometer. Results With the increased duration of heroin-abuse, T lymphocyte proliferation activity decreased significantly(P<0.05); T lymphocyte mitochondrial MPTP increased, △Ψm decreased, ROS increased(P<0.05). Sodium selenite can reverse these changes, and there is a dose-effect relationship(P<0.05). Conclusions Oxidative stress caused by prolonged heroin abuse has an adverse effect on the function and proliferation of T lymphocytes via the mitochondrial pathway. Sodium selenite has a protective effect on T lymphocytes by its antioxidant effect, which reduces the level of ROS to reverse the above process.
