目的 采用组织块贴壁法建立成人肠系膜动脉血管平滑肌细胞体外模型，为心血管疾病研究提供实验基础。方法 运用组织块贴壁法对成人肠系膜动脉平滑肌细胞进行原代培养，用倒置显微镜和免疫组化进行细胞形态学观察和鉴定。结果 运用组织块贴壁法成功获得大量单个成人肠系膜动脉平滑肌细胞，培养的细胞呈典型“峰－谷”样生长，免疫组化显示胞浆内平滑肌肌动蛋白阳性表达。经胰酶消化后可传20代。结论 通过组织块贴壁法可以获得成人动脉平滑肌细胞(human vascular smooth muscle cell，hVSMC)，为心血管疾病的实验研究提供良好的材料。

Objective To establish a method for culture of human mesenteric artery smooth muscle cell (hVSMC) by tissue-piece inoculation in vitro and to provide new technology to study cardiovascular diseases. Methods The tissue-piece inoculation method was applied to cell primary culture of hVSMC. Morphology of the cells was observed by inverted microscopy and identified by immunohistochemical methods. Results A great quantity of single hVSMC were obtained by tissue-piece inoculation. The cultured hVSMC showed typical “peak and valley” pattern and immunohistochemical staining showed that all the cells were positive. Conclusions The hVSMC are obtained by tissue-piece inoculation. This method provides good study material to research cardiovascular diseases.
