摘要

目的 研究α晶体蛋白对离体培养的嗅鞘细胞(olfactory ensheathing cells, OECs )存活和增殖的影响,探索增强移植OECs存活和增殖的有效方法。方法 取大鼠嗅球进行OECs离体培养,细胞计数检测α晶体蛋白组及对照组OECs的存活数目;Brdu掺入法及流式细胞仪检测OECs的增殖。结果 (1)培养5 d α晶体蛋白组OECs数目与对照组相比无明显差异,培养6~13 d,α晶体蛋白组OECs数目明显多于对照组(P<0.05);(2)将Brdu加入培养液中8 h后,α晶体蛋白组同视野Brdu阳性细胞数与总细胞数的百分比显著大于对照组(P<0.01);(3)α晶体蛋白组G2/M期、S期细胞数明显多于对照组(P<0.01)。结论 α晶体蛋白能明显促进离体培养的OECs的存活。

Abstract

Objective To investigate the effect of α-crystallin on the survival and proliferation of olfactory ensheating cells(OECs) in vitro. Methods OECs were cultured and treated with α-crystallin in DMEM/F12 or DMEM/F12 only. The number of surviving OECs was counte for after cultured 5days. The number of Brdu-labeled OECs was counted after Brdu was added in DMEM/F12. And cell cycle was analyzed by flow cytometry. Results α-crystallin promoted the survival of OECs at 6-13 days. The Brdu-labeled OECs in α-crystallin group was more in number than in control group. And the cell percentage of G2/M state and S state was higher in α-crystallin group than in control group. Conclusions α-crystallin promotes OECs survival and proliferation.
