目的 观察痹痛康丸对患胶原性关节炎(collagen-induced arthritis，CIA)大鼠血清肿瘤坏死因子α(TNF-α)的影响，探讨其治疗作用机制。方法 建立Ⅱ型胶原加完全弗氏佐剂诱导的大鼠CIA模型，60只Wistar大鼠随机分为6组，痹痛康丸低、中、高剂量组[0.3 g/(kg·d)、3 g/(kg·d)、30 g/(kg·d)]，尪痹冲剂组3 g/(kg·d)， 甲氨蝶呤片组每周1.2 mg/kg，采用药物溶解于2 ml生理盐水中，1次/d，连续灌胃给药4周，模型组灌服生理盐水2 ml/只。8周后处死大鼠，采用酶联免疫吸附法(enzyme-linked immunosorbent assay，ELISA)检测CIA大鼠血清TNF-α浓度，观察各组大鼠TNF-α浓度水平的变化。结果 痹痛康丸中剂量组TNF-α浓度水平(1190.84±177.02)、高剂量组TNF-α浓度水平(844.20±142.79)、MTX片组(1149.40±182.17)、尪痹冲剂组(1215.83±102.07)显著低于模型组(1724.67±247.26)(P=0.017， P=0.000， P=0.008， P=0.036)。痹痛康丸高剂量组血清TNF-α浓度水平显著低于痹痛康丸中剂量组、MTX片组、尪痹冲剂组，(P=0.006，P=0.018，P=0.009)。结论 痹痛康丸能有效降低CIA大鼠血清TNF-α的表达，有抑制CIA大鼠滑膜炎性的作用。

Objective To study the effect of Bitongkang Pills on TNF-α of collagen-induced arthritis (CIA) rats’ blood serum, explore its mechanism of therapeutic action. Methods The model of type II collagen with complete Freund’s adjuvant-induced rat CIA was established. 60 Wistar rat models were randomly divided into six groups, Bitongkang Pill low, medium and high dose groups [0.3, 3, 30 g/(kg·d)], costumes, Wang bi instant herbal medicine group 3 g/(kg·d) , methotrexate tablets group 1.2 mg/(kg·w) , the drugs of above groups were dissolved in 2 ml of saline, the rats were fed with the pills one time per day, intragastrically administered for 4 weeks continuously. The model group was fed with 2 ml saline each rat. After 8 weeks, the rats were sacrificed, underwent enzyme-linked immunosorbent assay (ELISA) to detect the CIA rat serum TNF-alpha concentration, and to observe each rat’s TNF-alpha concentration levels changes. Results The serum TNF-alpha levels in Bitongkang pills medium dose group (1190.84± 177.02), high-dose group (844.20±142.79), MTX tablets group (1149.40±182.17), Wangbi granules group (1215.83±102.07) were significantly lower than the model group (1724.67±247.26) (P=0.017, 0.000, 0.008, 0.036). The serum TNF-alpha levels in Bitongkang Pills high dose group were significantly lower than those in the Bitongkang Pills medium dose group, the MTX tablets group, and Wangbi granules group (P=0.006,0.018,0.009). Conclusions Bitongkang Pills can effectively reduce the CIA serum TNF-alpha expression, have the action of inhibiting CIA rat synovitis.
