目的 探讨肝癌细胞株Hep G2中凋亡抑制因子survivin与化疗药物敏感性的关系。方法 通过MTT检测肝癌细胞株Hep G2对于1.0 μg/ml顺铂, 1 μmol/L阿霉素,及1.0 μg/ml吉西他滨的敏感程度;通过Western blot 检测Hep G2中survivin 蛋白在顺铂、阿霉素、吉西他滨处理后不同时间段表达水平的变化。结果 在药物作用72 h后,10%~20%的肝癌细胞仍然存活并具有代谢活性。顺铂和吉西他滨处理48 h后survivin蛋白表达升高(2.04±0.05 vs 1.25±0.05;1.84±0.02 vs 1.25±0.05;1.92±0.05 vs 1.55±0.04;1.88±0.11 vs 1.55±0.04,差异有统计意义(P<0.05)。阿霉素处理48 h后survivin 蛋白表达并未见明显变化(1.73±0.11 vs 1.52±0.02;1.66±0.12 vs 1.52±0.02),差异无统计学意义。结论 survivin 蛋白水平升高与肝癌细胞株Hep G2部分化疗药物敏感性相关。
Abstract
Objective To investigate the relationship between survivin and chemotherapeutic sensitivity in hepatocellular carcinoma cells. Methods By the method of MTT, the sensitivity of Hep G2 was tested to be 1.0 μg/ml cisplatin, 1 μmol/L doxorubicin and 1.0 μg/ml gemcitabine. Western blot was used to detect the expression of survivin protein at various time points after the drugs intervention. Results Cultured for 72 hours, 10-20% of Hep G2 cells were still alive(2.039±0.05 vs 1.258±0.05;1.843±0.02 vs 1.258±0.05;1.924±0.05 vs 1.551±0.04;1.884±0.11 vs 1.551±0.04) ,P<0.05. After cisplatin and gemcitabine intervention for 48 hours, the expression of survivin was elevated (P<0.05). After adriamycin treatment for 48 hours, the expression of survivin protein had no significant change(1.729±0.11 vs 1.518±0.02;1.667±0.12 vs 1.518±0.02). Conclusions In Hep G2, the high expression of survivin is associated with the chemosensitivities of parts of drugs.
