eIF4E生物学功能及其在肿瘤中作用研究进展

摘要

随着分子生物学及肿瘤基因组学的研究进展,近年,针对肿瘤的分子靶向治疗进展迅速。肿瘤分子靶向治疗药众多,根据其作用机制可以分为两大类,一类是针对癌基因依赖靶点的靶向治疗[1],这类药物以引起肿瘤的驱动基因为靶点。另一类针对非癌基因依赖靶点的靶向治疗[2],这类药物的靶点蛋白在正常细胞中也存在,只是肿瘤细胞为了快速生长增殖对这类蛋白质需求量远大于正常细胞,例如针对分子伴侣热休克蛋白的靶向治疗。肿瘤细胞的生长增值需要大量的蛋白质,研究表明真核生物蛋白质调控相关蛋白异常与肿瘤发生发展有关,因此针对蛋白质翻译调控靶点成为非癌基因依赖靶向治疗的研究热点。而真核细胞蛋白质翻译调控的限速步骤主要在翻译起始阶段,而真核细胞翻译起始因子4E(eukaryotic traslation initiation factor 4E, eIF4E)作为翻译起始阶段的重要调控因子在多种肿瘤中表达异常。本文就eIF4E在肿瘤中的作用及以其作为靶点的分子靶向治疗研究现状给予综述。
