摘要

目的 探讨丹参治疗大鼠口腔黏膜下纤维化的效果。方法 选取8周龄SPF级雄性健康Sprague-Dawley(SD)大鼠70只,采取槟榔碱涂擦法建立SD大鼠口腔黏膜下纤维化模型后,将大鼠分为6个实验组和对照组,每组10只。实验组分别局部注射5 mg/ml丹参酮ⅡA、丹参酮ⅡB、隐丹参酮、异隐丹参酮、丹酚酸A、丹酚酸B,对照组局部注射生理盐水,处理8周后测量局部黏膜病变面积后取局部颊黏膜行HE染色观察病理变化;并检测组织内Ⅲ型胶原、TGF-β1和IFN-γ的表达情况。结果 丹参酮ⅡA、隐丹参酮和异隐丹参酮组的口腔黏膜下纤维化面积减小分别达到2.72、1.95、1.58 cm2,治愈率60%~80%,明显优于对照组,差异有统计学意义(P<0.05)。丹参酮ⅡA、隐丹参酮和异隐丹参酮组的Ⅲ型胶原蛋白灰度比值、TGF-β1蛋白降低明显,IFN-γ蛋白明显增高,与对照组相比差异有统计学意义(P<0.05);而丹参酮ⅡB、丹酚酸A和丹酚酸B组的黏膜病变减少量、治愈率及Ⅲ型胶原、TGF-β1和IFN-γ蛋白的表达与对照组比较,差异均无统计学意义。结论 丹参酮ⅡA、隐丹参酮和异隐丹参酮具有治疗口腔黏膜下纤维化的效果。

Abstract

Objective To study the therapeutic effect of salvia miltiorrhiza on oral submucosal fibrosis in rats. Methods After the establishment of the SD rat OSF model using arecoline smear method, the rats were divided into 6 experimental groups and 1 control group. The experimental group received 5 mg/ml tanshinone ⅡA, tanshinone ⅡB, cryptotanshinone, isocryptotanshinone, salvianolic acid A, and salvianolic acid B, while the control group were treated with physiological saline. After 8 weeks of treatment, the local mucosal lesion area was measured and the local buccal mucosa was stained with HE to observe the pathological changes. The expressions of type Ⅲ collagen, TGF-β1 and IFN-γ were detected. Results The area of oral submucosal fibrosis in the tanshinone ⅡA, cryptotanshinone and isocryptotanshinone groups decareased by 2.72, 1.95, and 1.58 cm2, respectively, and the cure rate was 60%-80%, which was statistically significant compared to the control group.The type Ⅲ collagen in the tanshinone ⅡA, cryptotanshinone, and iso cryptotanshinone groups decreased by 0.06, 0.04, 0.03,TGF-β1 decreased by 0.08, 0.04, 0.05, and IFN-γ increased by 0.06, 0.03, 0.03, with statistically significant differences compared to the control group (P<0.05).There was no statistically significant difference between the tanshinone ⅡB, salvianolic acid A, and salvianolic acid B groups and the control group. Conclusions Tanshinone ⅡA, Cryptotanshinone and Isocryptotanshinone have therapeutic effects on OSF in rats, while Tanshinone ⅡB, salvianolic acid A and salvianolic acid B do not have therapeutic effects.
