半夏泻心汤加减方含药血清活性成分分析及体外抑菌作用

半夏泻心汤加减方含药血清活性成分分析及体外抑菌作用

摘要

目的 分析半夏泻心汤加减方含药血清中的主要活性成分及其体外抑菌作用。方法 将半夏泻心汤加减方熬制浓缩后,大鼠灌胃制备半夏泻心汤加减方含药血清,高效液相色谱-串联质谱(HPLC-MS-MS)法测定血清中主要药物活性成分。根据HPLC-MS-MS结果,通过微量肉汤稀释度法体外抑菌实验,检测半夏泻心汤加减方主要活性成分和含药血清对幽门螺杆菌及其他细菌抑制作用。结果 检测结果显示,含药血清中的主要活性成分含量由大到小依次为槲皮激素、异鼠李素、黄芩苷、琥珀酸、党参炔苷。体外抑菌实验结果显示,党参炔苷抑制细菌(金黄色葡萄球菌、白色葡萄球菌、大肠杆菌、沙门菌、变形杆菌、幽门螺杆菌)的最低抑菌浓度(MIC)为100 μg/ml,对异鼠李素抑制幽门螺杆菌MIC为400 μg/ml,对其余细菌为200 μg/ml;黄芩苷抑制幽门螺杆菌的MIC为400 μg/ml,对其余细菌为200 μg/ml;槲皮素MIC为200 μg/ml。含药血清按照体积比MIC为1∶4(金黄色葡萄球菌、大肠杆菌和变形杆菌),1∶1(沙门菌和幽门螺杆菌);琥珀酸则不抑制这些细菌的生长。结论 半夏泻心汤加减方中含药血清主要活性成分党参炔苷、黄芩苷、槲皮素及异鼠李素具有抑制幽门螺杆菌和其他细菌的作用,阐明了半夏泻心汤加减方抑菌抗炎的主要原因。

Abstract

Objective To analyze the main active components in the serum of Banxia Xiexin decoction were analyzed,and its antibacterial effect in vitro. Methods After decocting and concentrating Banxia Xiexin decoction,the drug-containing serum of Banxia Xiexin decoction was prepared by intragastric administration to rats. The main drug active components in the serum were determined by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS-MS). According to the results of HPLC-MS-MS results,the inhibitory effects of main active components and drug-containing serum of Banxia Xiexin decoction on Helicobacter pylori and other bacteria were detected by microbroth dilution method in vitro. Results The test results showed that the contents of the main active components in the drug-containing serum, from high to low, were quercetin, isorhamnetin, baicalin, succinic acid, and codonopsis glycoside. The results of the in vitro antibacterial experiments showed that minimum inhibitory concentration (MIC) of codonopsis pilosula for inhibiting bacteria (staphylococcus aureus, staphylococcus albicans, escherichia coli, salmonella, proteus, helicobacter pylori) was 100μg/ml.,the MIC of isorhamulin was 400 μg/ml for helicobacter pylori,and the remaining bacteria were 200 μg/ml. The MIC of baicalin in helicobacter pylori was 400 μg/ml,and others were 200 μg/ml. The MIC of quercetin was 200 μg/ml. The MIC of drug containing serum was in a volume ratio of 1:4 (staphylococcus aureus,escherichia coli and proteus) and 1:1 (salmonella and helicobacter pylori). Succinic acid did not inhibit the growth of these bacteria. Conclusion The main active components of the drug-containing serum in the modified formula of Banxia Xiexin decoction, namely codonopsis glycoside,baicalin,quercetin and isorhamnine,have the effect of inhibiting helicobacter pylori and other bacteria, clarifying the main reason the main reasons for the antibacterial and anti-inflammatory effects of the modified formula of Banxia Xiexin decoction.
